什么是Phos-tag™
Phos-tag ™ 是一种能与磷酸离子特异性结合的功能性分子,结合特异性与氨基酸的种类和序列无关,无放射性,无需特意制备磷酸化抗体。它可用于磷酸化蛋白的分离(Phos-tag™ Acrylamide)、Western Blot 检测(Phos-tag ™ Biotin)、蛋白纯化 (Phos-tag ™ Agarose)及质谱分析MALDI-TOF/MS (Phos-tag ™ Mass Analytical Kit) 。

1. Phos-tag™ Acrylamide

Phos-tag Acrylamide SDS-PAGE可根据迁移率不同,区分磷酸化蛋白与非磷酸化蛋白。该方法只需在常规的SDS-PAGE胶中添加Phos-tag Acrylamide和MnCl2即可进行实验(制胶过程中添加,如需详细说明请联系我们)。

原理

应用


1、黄色粘性油状物质;
2、在4度可保存至少一年;
3、Phos-tagTM与丙烯酰胺(Acrylamide,SDS-PAGE分离胶主要成分)结合分离磷酸化和非磷酸化蛋白。
SDS-PAGE分离胶制备中添加金属离子(Zn2+或者Mn2+)和Phos-tagTM Acrylamide。
在蛋白电泳过程中,磷酸化基团与金属离子螯合的Phos-tagTM Acrylamide发生特异性结合,导致磷酸化蛋白的迁移速度变慢。
Phos-tag® Acrylamide可根据迁移率不同,区分磷酸化蛋白与非磷酸化蛋白。无放射性、非化学物质标记。不同位点磷酸化修饰的蛋白在同一泳道中因迁移率不同而被分离开来。
操作过程与普通SDS-PAGE相类似。Phos-tagTM的特异性结合与氨基酸种类、序列无关。
适用于免疫印迹、质谱分析等后续工作。可识别磷酸化和非磷酸化蛋白。
Phos-tagTM母液可稳定保存至少3个月。
可检测出具有不同磷酸基团数目的蛋白。无需特意制备磷酸化抗体。
